Another Day in Paradise
The scene in the woods with James Woods and Vincent Kartheiser was completely improvised and involved Woods hitting Kartheiser repeatedly with his fingers. The gestures are so rough and sudden that you can hear each hit and see Kartheiser's genuine surprise, respectively. Afterwards, Kartheiser went up to director Larry Clark and said, simply, "I didn't know that motherfucker was going to hit me."
Beetle Juice
Originally planned as a supernatural drama entitled "The Maitlands" about a deceased couple dealing with the troubles of the afterlife. The scenes were scripted much more morbidly than they appear (most notably the waiting room scenes, which were supposed to be particularly ghoulish), and most of the characters were much darker. Only when Michael Keaton began filming his scenes and asked for creative freedom did Tim Burton realize the potential of a dark comedy

In a interview, Stacy Ferguson stated that during her scenes with Quentin Tarantino, he had bitten her on the neck. She said that it was one of the coolest moments filming, despite her agent almost wanting to sue Tarantino for that stunt.

Serenity
According to Joss Whedon in his commentary, the most difficult scene to get the sound right was the scene between Simon and River at the end, after Simon had been shot. Whedon says that the bond formed between Sean Maher and Summer Glau over the course of the shooting of Firefly was so strong, and Sean's acting so good, that Summer kept bursting into tears whenever they attempted to shoot the scene. (Poster's note: Awww. Sean/Summer RPF, anyone?)
Sin City
Because of the way the movie was shot, Mickey Rourke (Marv) and Elijah Wood (Kevin) never met until after the film was released. (Poster's note: For those of you who have never seen this movie, Marv and Kevin have two fight scenes, and Marv later tortures Kevin. There's also an extended portion of the story where the two are handcuffed together. So this is pretty damn funny and gives you an idea of just how much CGI the film used.)
Star Wars - Episode IV: A New Hope
Carrie Fisher's breasts were taped down with gaffer tape as George Lucas had not considered the possibility of people wearing underwear in space.
Troy
Brad Pitt and Eric Bana both did their own fighting scenes. They also made an agreement that they would pay for every hit they accidentally made. The costs were $50 for a lighter blow and $100 for a hard blow. Pitt ended up paying $750 to Bana, who didn't have to pay Pitt anything.
